So it shows that it can't find some file. The problem is that usually this error is more detailed and shows the file path...
Try to close
Kindle for PC before running DRM removal tools.
If this doesn't work, try to install
Kindle for PC 1.6.1, delete all Kindle ebooks and re-download them. Then try with to use the
Calibre plugin again.

You may find it in the following folder:
<User home>\...\Amazon\Kindle For PC\{AMAwzsaPaaZAzmZzZQzgZCAkZ3AjA_AY}
where ... is
- "Local Settings\Application Data" in Windows XP
- "USERS/username/AppData/" in Windows 7
